Reagents Volume ml Protection buffer 100 Lysis buffer 20 Wash buffer I 30 Wash buffer II 30 Elution buffer 5 Magnetic Beads 2 Isopropanol 15 β-Mercacptoethanol or DTT 1 1, collect 1-2 ml saliva and add equal protection buffer into the saliva, and centrifuge it at 2000 RPM for 1 minute. Discard supernatant. About 50-100 µl residue pallet for next extraction. 2, Add 200 µl lysis buffer into the residue pallet and mix well. Add 20 µl magnetic beads (25mg/ml) and 10 µl β-Mercacptoethanol or DTT (1M), 200 µl isopropanol into the mixture. Mix them very well at room temperature for 2 min. 3, Set the tube on the magnet for 20 seconds. Remove the supernatant. 4, Add 300µl wash buffer I and vortex it. Set it up on the magnet rack and remove the supernatant. 5, Add 300 µl wash buffer II and vortex it. Set it up on the magnet rack and remove the supernatant. 6, Add 200 µl 0 ℃ cold water gently and quickly wash it. Set it on the magnet rack and rapidly remove the supernatant. 7, Add 50 µl elution buffer and vortex it well. Set it at 50 ℃ for 5 min. Set it on the magnetic rack and collect about 50µl supernatant. Save it in -80 ℃ for next use.
